Genetically engineered derivatives of lambda DNA are used as cloning vectors. The DNA is frequently used as substrate for restriction enzyme characterization and activity assays. Lambda DNA is isolated as a double-stranded linear molecule and contains 48,502 base pairs. The complete genome nucleotide sequence was first sequenced in 1982.

The DNA possesses 12 base single-stranded termini that are complimentary. During replication in the infected cell, these termini anneal together and are sealed to form a circular lambda DNA molecule. The DNA is converted into a linear molecule during packaging into viral particles.
